Drug intelligence / Profile preview
hyperpolarised [1-13C] pyruvate is an investigational diagnostic imaging agent used in combination with carbon-13 ($^{13}\text{C}$) magnetic resonance imaging (MRI) and magnetic resonance spectroscopic imaging (MRSI) to non-invasively visualize and quantify real-time cellular metabolism in vivo. Developed initially by GE Healthcare, the agent is hyperpolarized using dissolution dynamic nuclear polarization (dDNP), which temporarily increases the nuclear spin polarization of the $^{13}\text{C}$ nuclei by over 10,000-fold, dramatically enhancing the signal-to-noise ratio. Following intravenous injection, the metabolic flux of the hyperpolarized pyruvate is tracked as it is enzymatically converted into downstream metabolites, primarily $^{13}\text{C}$-lactate (via lactate dehydrogenase), $^{13}\text{C}$-alanine (via alanine transaminase), and $^{13}\text{C}$-bicarbonate (via pyruvate dehydrogenase). This metabolic profiling is utilized to detect and characterize altered metabolic states (such as the Warburg effect in aggressive cancers like prostate and pancreatic cancer, or altered myocardial metabolism in heart failure) and to monitor early therapeutic responses.
